بررسی امکان نگهداری بذر گونه Eucalyptus microtheca در شرایط فراسرد (Cryopreservation)

چکیده

به‌منظور بررسی امکان نگهداری بذر گونه Eucalyptus microtheca در شرایط فراسرد، از پیش‌تیمارهای گلیسرول 30 درصد، محلول ویتریفیکاسیون گیاهی (Plant Vitrification Solution 2 یا PVS2) و کاهش رطوبت بذر (Desiccation) استفاده شد. بذرهای پیش‌تیمار شده به‌مدت یک هفته در ازت مایع با دمای Cº 196- نگهداری شدند و پس از خروج از ازت مایع در معرض شوک حرارتی (Cº 42+) قرار گرفتند. پس از اعمال شوک حرارتی نیمی از بذرها بر روی کاغذ مرطوب کشت و به ژرمیناتور با دمای Cº 24+ منتقل شدند. نیمی دیگر درون گلدان حاوی خاک معمولی و پیت‌‌ماس کشت و به گلخانه با دمای   Cº 20+ انتقال یافتند. بذر تیمارهای مختلف که مرحله ماندگاری را در شرایط فراسرد گذرانده بودند، ضمن احیا مجدد قادر به جوانه‌زنی و تولید گیاهچه بودند. بذرهای تیمارهای مختلف فراسرد که در شرایط ژرمیناتور و گلخانه رشد کردند، تفاوت معنی‌داری با بذرهای شاهد نداشتند و رشد گیاهچه‌ها طبیعی و عاری از هر گونه آثار سوء بود. نتایج حاصل از این بررسی نشان داد که با توجه به کاهش شدید فعالیت‌های متابولیک و فیزیولوژیک موجود زنده در این دما، امکان نگهداری بلندمدت بذر گونه E. microtheca در شرایط فراسرد کاملاً امکان پذیر بوده و می‌توان انتظار داشت که بذرهای این گونه را بتوان با استفاده از تکنولوژی فراسرد برای مدت زمان بسیار طولانی حفظ نمود

چکیده [English]

The aim of this study was to investigate the possibility of cryopreservation of Eucalyptus microtheca seeds at -196 ºC, using Liquid Nitrogen (LN). Before transferring the seeds into LN, three pre-treatments including Plant Vitrification Solution 2 (PVS2), Desiccation and Glycerol 30% was applied. The treated seeds transferred into LN for period of one week. The seeds removed from the LN, subjected to heat shock (+42 ºC) for 2 minutes, blotted and transferred both onto moist paper in petri dishes and pots, filled with soil and pit moss. The petri dishes and pots, transferred into germinator (+24 ºC) or greenhouse (+20ºC), respectively. Subsequent of cryopreservation period, seed germination and recovery was high, there were no significant differences between cryopreserved and control seeds in germinator and greenhouse conditions. Further more, there were no adverse effect or abnormality observed in seedlings developed from cryopreserved seeds. Results showed that, long-term preservation of E. microtheca seeds under LN (cryopreservation) is promising.
