Seem. is an important evergreen plant species from different points of view. This species has important characteristics including resistance to high drought and temperature, sand fixation, medicinal properties, and durable wood. The objective of this study was to investigate the genetic diversity among 30 genotypes of the species collected from Bushehr, Kerman and Hormozgan provinces using molecular data obtained from SSR primers. DNA samples were extracted and used in PCR tests by applying microsatellite primers. After agarose gel electrophoresis of DNA Fragments, a totally 29 alleles were distinguished. In831 primer generated most locus. Results of cluster analysis using the UPGMA algorithm based on the Jaccard similarity coefficient categorized the genotypes into eight groups, indicating a high level of genetic diversity among the genotypes. Furthermore, observed and expected heterozygosity were estimated to be 0.4 and 0.29, respectively. Furthermore, the effective number of alleles ranged between 1.2 in the In831 gene to 1.81 in the Tau16 gene with average values of 1.51. These results showed that SSR markers are highly effective in determining the amount of genetic diversity in the population of T. undulata. The observed high diversity among the genotypes of the species showed a rich source of germplasm which can be used in breeding programs.}, keywords = {Genetic diversity,genotype,germplasm,primer}, title_fa = {بررسی تنوع ‌ژنتیکی انار‌شیطان (Tecomella undulata (Roxb.) Seem.) ‌‌ویژگی‌‌هایی مهمی مانند مقاومت مناسب به دمای زیاد و خشکی، تثبیت شن‌‌های روان، خاصیت دارویی و نیز چوب محکم و بادوام ‌‌دارد. در این پژوهش، تنوع ژنتیکی 30 ژنوتیپ از این گونه در استان‌‌های بوشهر، کرمان و هرمزگان با استفاده از نشانگر SSR بررسی شد. پس از استخراج DNA، تکثیر با استفاده از چهار آغازگر ریزماهواره به‎روش PCR انجام شد. پس از الکتروفورز قطعات تکثیرشده در ژل آگارز، در‌مجموع 29 مکان ژنی شناسایی شد. آغازگر In831 بیشترین تعداد مکان (15) را تولید کرد. ماتریس تشابه داده‌های اثر انگشت جدایه‌ها در SSR با محاسبه ضریب جاکارد ترسیم و بررسی شد. دندروگرام‌‌ها نیز با روش میانگین‌های حسابی بی‌‌وزن (UPGMA) ترسیم شدند. براساس نتایج، ژنوتیپ‌‌ها به هشت گروه تقسیم شدند که بیانگر تنوع ژنتیکی زیاد ژنوتیپ‌‌ها در این پژوهش بود. در این بررسی، میانگین هتروزیگوتی مشاهده‌شده و موردانتظار به‌ترتیب 0/4 و 0/29 برآورد شد. همچنین، تعداد آلل مؤثر از 1/2 در مکان ژنی In831 تا 1/81 در مکان ژنی Tau16 با میانگین 1/51 متغیر بود. این نتایج نشان می‌‌دهد که نشانگرهای SSR در شناخت اندازه تنوع ژنتیکی در جامعه گیاهی انارشیطان، کارایی زیادی دارند. تنوع ژنتیکی زیاد بیانگر منبع غنی ژرم‌‌‌پلاسم در این گیاه است که از آن می‌‌توان در برنامه‌‌های به‌‌نژادی استفاده کرد.}, keywords_fa = {آغازگر,تنوع ژنتیکی,ژنوتیپ‌‌,ژرم‌‌پلاسم}, url = {https://ijfpr.areeo.ac.ir/article_120121.html}, eprint = {https://ijfpr.areeo.ac.ir/article_120121_1d9c9a7a481f9becca0bf3d987416ad6.pdf} }
